神亀5年に越智玉純がこの地を霊地と悟り熊野十二社権現を祀った。天平元年に行基が薬師如来を刻んで本尊として安置し開基した。弘仁4年に空海が訪れて真言宗に改められ、後に衛門三郎再来の伝説により石手寺と改められた。
